Project summary The lectins are carbohydrate binding proteins that agglutinate cells or precipitate glycoconjugates. Lectins exhibit diverse functions like mitogenicity, immunomodulation, antimicrobial activity, antiviral and antiproliferative properties. In cancer research lectins are extensively used as diagnostic tool and also as antiproliferative agent. In the present study, Con A lectin is used to study the genes involved in mitogenicity and antiproliferative effect induced by this lectin. Microarray technique is used to study the gene expression induced by Con A in normal human normal lymphocytes and acute myeloid leukaemia cell line (HL 60). Introduction Origin of the proposal • Binding of mitogenic lectins -involving different signal transduction pathways which ultimately results in cell proliferation. • Lectins have been found to possess anti-cancer properties • Concanavalin A is a mannose/glucose binding lectin isolated from Canavaliaensiformis. • Con A act as a T-cell mitogen and in cancer cells it induces cell death through apoptotic and autophagicpathway different molecular processes involved in mitogenicityand cytotoxicity • Gene expression study • Microarray
Definition of the problem To study the genes expression induced by concanavalin A in human lymphocytes and cancer cells using microarray.
Objectives • To optimize the concentration of Con A for mitogenicity and anti-proliferative effect in human lymphocytes and cancer cells. • To study gene expression in untreated lymphoctes and Con A treated lymphocytes using microarray. • To study gene expression in untreated cancer cells and Con A treated cancer cells using microarray.
Review of status of Research and Development in the subject International status • Con A enhanced osteogenesis in human bone marrow MSC- osteocalcin, RUNX2, BMP-2, BMP-4, and BMP-6 mRNA expression (Sekiya K et al., 2008) • Increased in telomere length and replicative capacity of peripheral blood mononuclear cells (Murillo- Ortiz B et al., 2013) • Induced apoptosis by p73 regulated Akt-Foxo1a-Bim pathway in p53 deficient cells (Amin AR et al., 2007) • ConAand Sophoraflavescenslectin (SFL) induces apoptosis in Human breast carcinoma MCF-7 cells through p53 dependent pathway by reducing NF-κB, ERK, and JNK levels, and increasing p53 and p21 levels (Shi Z et al., 2014) • Con A induced BNIP3-mediated mitochondrial autophagy in hepatomacells(Chang CP et al., 2007) National status Con A induced autophagy through MEK/Extracellular signal-regulated kinases (ERK) pathway in Human cervical cancer (HeLa) cells was studied by IIT Kharagpur (Roy B et al., 2014)
Novelty Importance of the proposed project • Con A induced gene expression study using microarray technique • Since this genomic study compares the normal with the cancerous cells, it will give the differential pattern in expression gene which is basis for tumorigenesis. Such genes can be used as a therapeutic marker in diagnosis of cancer.
Target beneficiaries of the proposed work • This in vitro study can be translated to clinical practice in patient tumor samples to analyse the biological markers for an early detection, monitoring, prognosis of the disease and response to therapy.
Methodology: • Materials: • Method: • To optimize the concentration of Con A for mitogenicity and anti-proliferative effect in human lymphocytes and cancer cells • Mitogenic potential of Con A is determined against human lymphocytes as described by Singh RS et al. • Antiproliferative effect Con A on cancer cells(HL-60 Cell line) is studied using MTT assay (Yan Q et al., 2009) • To study gene expression in lymphocytes and cancer cells using microarray (Marimuthu A et al., 2011) • Sample goups: untreated lymphoctes and Con A treated lymphocytes • untreated cancer cells and Con A treated cancer cells( HL-60)
The development "Outcomes" and "Outputs' of the project • Research publications in high impact journals. • This study opens multidisciplinary research areas like genomics, proteomics, Cancer research for future studies • NCBI database can be updated by adding the microarray data obtained from the study. • The microarray data can be used to detect a biomarker for an early detection, monitoring, prognosis of the disease and response to therapy.
