About Microbiology Microbiology is simply the study of microscopic organisms that are in every place thought imaginable Crucial and important to gain information and understand microbial diversity in order to find the origins of how viruses and molecular diseases develop To be a microbiologist, one must be curious about small microscopic life, study microorganisms from various areas, and be rigorous in the study of microbiology
Procedures and Techniques Inoculation Culturing Isolation FISH (Fluorescence in Situ Hybridization)
Results From Culturing in Petri Dishes After (Incubation #2 After Isolating One Colony) Before (Incubation #1) (collected from the Memorial Pond)
Results continued… White cells stick together One spot of separation in colonies Single colony in the middle of agar dish was the same color Before (Incubation #1) After (Incubation #2 After Isolating One Colony) (collected from the bottom of a shoe)
More results… Rough edges with wrinkled white pinches in the center Off-white color and cells clumped together Wrinkled pinch has a rotational symmetry design After (Incubation #2 After Isolating One Colony) Before (Incubation #1) (collected from refrigerator)
Microscopy Results All samples had unviersal bacteria There were no eukaryotes in any of the samples All samples contained Gama-proteobacteria Marianelly and Edima’s samples contained Archaea and Actinobacteria as well
FISH Pictures • Marianelly: Red, universal Bacteria; yellow (orange) and green, Gama Proteobacteria; blue, Archaea and Actinobacteria.
FISH Pictures Continued… • Edima: Red, universal Bacteria; yellow, Gama Proteobacteria; blue, Archaea and Actinobacteria.
FISH Pictures Continued… • Kelechi: Red, universal Bacteria; yellow (orange) and green, Gama Proteobacteria.*
Conclusions We were capable of discovering microbial variety in our chosen environmental samples. In addition, we could grow microorganisms in the lab. Lastly, we were successful in identifying organisms within the Bacteria and Archaea domains. Overall, our experiment was a success.
