Post-column Derivatization • Renders visible certain compounds that are normally invisible by performing a chemical reaction on the substances that gives them an easily detectable physical property • Reaction typically produces a strong color or fluorescent product • Used to increase sensitivity and selectivity in HPLC analysis • Mixes the stream of effluent flowing from the HPLC column with a stream of reagent solution • Compounds and reagent flow into reaction chamber • Typically heated • Some methods require two (or even three) reactions in series • Derivatized compounds pass into a detector
