Metrics, Milestones, and Timelines PSI Target Selection Meeting Stephen K. Burley November 14th 2003
Agenda • Status quo • Examples taken from PSI-funded center websites • Rationale for adopting a common system for presenting Metrics • Proposed common system for presenting Metrics (i.e., PX and NMR straw men) • Center responses to the straw men • Rationale for making clear declarations of both Milestones and Timelines
Status Quo • Metrics • NIGMS-mandated declaration of aggregate numbers by each PSI-funded center, but there is considerable variation in what is presented and how it is presented. • Milestones • Essentially no information. • Timelines • Essentially no information.
“Problems” with Current Presentations of Metrics • Significant differences in format make it difficult to impossible to find common statistics • Significant differences in level of detail (Predictably, the most numerically successful centers are the most communicative!) • Complete absence of quarterly productivity statistics (hurts centers that are ramping up) • Incomplete information on process stage dwell time for each target (makes internal prioritization difficult to impossible)
Rationale for Common Presentation of Metrics • To provide critical cumulative information regarding progress to date in the same format on every PSI center website • To provide the same statistics for each quarter • To provide a means of measuring how long a particular target spends in a particular phase of the process • To make the lives of the PSI PIs • (and John Norvell!) easier
Proposed Common Presentation of Metrics I • PX Cumulative and Quarterly (effective 9/1/03) • - # target genes cloned • - # soluble targets • - # targets purified • # targets in crystallization trials • # targets abandoned • # targets giving crystals that diffract (<3Å) • # target structures phased • # target structures refined • # target structuresPDB (identify new fold)
Proposed Common Presentation of Metrics II • NMR Cumulative and Quarterly (effective 9/1/03) • - # target genes cloned • - # soluble targets • - # targets purified • # targets screened by HSQC • # targets abandoned • # targets with backbone assignments • # targets with sidechain assignments • # target structures refined • # target structuresPDB (identify new fold)
Discussion • MCSG-A Joachimiak • JCSG-RC Stevens • BCSG-SH Kim • TBSGC-DC Eisenberg • SESGC-BC Wang • SGPP-WGJ Hol • CESG-JL Markley • NESGX-GT Montelione
Presentation of Milestones/Timelines • To provide a means of measuring progress versus goals • To quantify bottlenecks and make rational choices regarding investments in automation, etc. • To identify languishing targets and make objective decisions about whether or not and how to continue • To make John Norvell’s life easier
Multiple Organisms Ensure Soluble Protein Species Af to Tm • 40 Species • 262 Gene families • 1852 Gene sets • Soluble protein(s) for 80% of gene families Gene number Summary
