  1. Genomic Prep: Did it work?

  2. Is it DNA? • Ways to Tell: • Microscope? Telescope? • Some Dye? • Spectroscopy • Gel

  3. Spectroscopy • How it works • Absorbance • DNA- 260nm • Proteins- 280nm

  4. What is Purity? • What does dirty DNA mean? • Is there actual “dirt” on it? • Purity also refers to the presence of RNA and Protein. • Purity: Absorbance of DNA (260nm)/Absorbance of Protein (280nm) • Super important info: • An optical density (or "OD") of 1 corresponds to a concentration of 50 μg/ml for double-stranded DNA.

  5. What is a good Purity ratio? • Guesses? • What does a ratio greater than ONE mean? Less than ONE? • Acceptable purity ratio: greater than 1.8 • Meaning that the DNA is not contaminated protein.

  6. Gel • What would a Gel look like if only Genomic DNA and dye were added?

  7. Example From a Draft 2 paper
