Gel Electrophoresis DNA Unit
Rationale • Present the topics of cell function, DNA and the applications of DNA analysis in science • DNA is a defined fingerprint to each individual human • Introduce the students to occupations that use DNA analysis
Learning Outcomes • Students will label the different parts of the cell and define cell functions. • Students will analyze the structure of DNA; then describe how the body uses DNA. • Students will learn compare gel electrophoresis to a variety of DNA analysis and research how to use it DNA analysis in modern science.
Saskatchewan Objectives • To enable students to understand and use the vocabulary, structure and forms of expression which characterize the study of biology. • To develop a contemporary view of technology. • To develop compassionate, empathetic and fair-minded students who can make positive contributions to society as individuals and as members of groups.
Topic Sequence Cells & Their Function DNA & Replication GelElectrophoresis
Cells and their Function Main Points • Plants vs.Animal cells • Parts of the cells(plant and animal) then compare the two • Functions of the cell and the functions of the cell parts • Grade Dependant: • Incorporate photosynthesis and Calvin Cycle • Photosynthesis- light and dark reactions • Metabolism- chemical pathways to produce ATP
DNA Replication Main Points: • DNA is unique to each individual • Hereditary – light introduction • Where DNA replication takes place • What parts of the cell participate • Types of RNA • Codon and Anti codon • Proteins
Gel Electrophoresis • Main Points: • DNA is denatured through boiling and DTT • The proteins that are analyzed • Voltage pushes the proteins down the gel • Uniform charge of the proteins from the SDS • Proteins separated by size • 4-20% gels • Gels should be ran on 120 volts for 1 hr or 90 volts for 1 ½ hrs… to high of voltage results in melted gels
Assessment • Hand outs - draw and label the cell - functions of the cell • Make a movie , audio or storyboard of DNA replication • Starting with a broad perspective use your vocabulary to focus the topic to how DNA effects humanity. • Gel electrophoresis lab • Lab notebooks – write a lab • Research uses of gel electrophoresis • Exam
