Lab 1: Chemistry, Pipetting, and Biuret’s Test Dr. Sarah VanVickle-Chavez BIO 2960 Laboratory Spring 2013
Chemistry for Biology Review • Use Firefox or Safari (Internet Explorer doesn’t always cooperate) • http://www.nslc.wustl.edu/courses/Bio2960/labs.html • There will not be a lab report due next week. But…. • There are three reports due at the beginning of class in two weeks: • 1a = proteins (we’ll do this next week) • 1b = cell organelles (use your book/notes for this one) • 2 = SDS-PAGE (we’ll do this next week)
Pipetting • P-20 1-20 ± 0.5μl • P-200 20-200 ± 1 μl • P-1000 200-1000 ± 10 μl • The smaller pipettors are always more accurate. • The top number on the dial should be the top number in your volume. Demo: http://www.youtube.com/watch?v=cxm61exlVnc
Biuret’s Test Remember: Cu2+ = BLUE, but a positive rxn is VIOLET. Why? Biuret’s = CuSO4 + NaOH (to keep pH basic) If there’s a peptide bond, Cu2+ complexes w/ 4 Ns. The more bonds, the more complexes. Additional Notes: http://www.youtube.com/watch?v=wtVGM6tIhHg http://www.piercenet.com/browse.cfm?fldID=876562B0-5056-8A76-4E0C-B764EAB3A339
