MICROBIOLOGY LABWORKJPEMS 2012NANTES Dr C. Bressollette (Virology, Nantes) Dr J. Caillon (Bacteriology, Nantes) Dr M. Eveillard (Bacteriology, Angers) Cécile Peltier (Virology, Nantes)
The week schedule • Virology • Viral cultures: • Observation of a cytopathic effect • HSV typing by immunofluorescence • Molecular assay • Amplification of a viral gene • Serologic assay: • Inhibition of hemagglutination • Bacteriology • Analysis of a bacterial mixture • Gram staining • Bacterial identification • Susceptibility to antibiotics • Analysis of the environment
Day 2 – Viral cultures • Observation of non infected adherent cells under the microscope Human fibroblasts Primary cells, limited multiplication in vitro HEP-2: epithelial cells, immortalized cell line
Day 2 – Viral cultures • Observation of infected adherent cells under the microscope Enterovirus Respiratory Syncitial Virus
Day 2 – Viral cultures • Observation of infected adherent cells under the microscope HSV Adenovirus
Day 2 - Virology • Observation of infected adherent cells under the microscope Cytomegalovirus
Day 2 - Bacteriology • Analysis of the bacterial mixA, B, C, D ,E : Observe the aspect of the colonies on the 3 agar plates • Chromogenic agar plate : 2 sorts of colonies • Chapman : only 1 type of colonies • Drigalski : only 1 type of colonies • lactose positive (yellow colonies) • or lactose negative (blue-green colonies) according to the student pairs
Day 2 - Bacteriology • Biochemical identification of the colonies isolated on the Drigalski agar plate • Seed an identification gallery • Study the biochemical characters of the bacteria • See the demonstration and the lab work workbook
Bacterial identification Api system Scratch 2 or 3 colonies from the agar plate Rub on the glass wall and moisten progressively Sterile distilled water Not underlined Not framed Just fill the tubule with the suspension Cup Tubule Underlined Cover with oil in the cup Framed: fill completely (tubule + cup)
Antibiogram by disk diffusion Principle the antibiotic disk gets wet in contact with the agar It diffuses into the gel, with a decreasing gradient of concentration from the disk to the periphery. Method • - Prepare a bacterial suspension from a colony with a sterile loopful, in a tube of sterile distilled water; • Dip the swab in the suspension • Inoculate the agar plate with close streaks,
Antibiogram by disk diffusion Method Place the antibiotic disks immediately.
