Work package 7 Establishing new product/processes for bioremediation Partners: • UMIL (2 PM) • IAMAC (11 PM) • UGENT (26 PM) – WP Leader • ETS (19 PM) • UNIBO (34 PM)
Objectives Development of biotechnological processes based on microorganisms from the collections (WP4): • the biosynthesis of valuable fine chemicals and new biosurfactantsby hydrocarbon degrading microbe • the reductive dehalogenation of chlorinated compounds with the new isolates • the reductive dehalogenation of PCBs by biopalladium • biotransformation of metals and metalloids in polluted sediments and definition of models describing their fate
Task 7.1 - hydrocarbon degradation enzymes and microorganisms to obtain valuable chemicals – focus on biosurfactant • isolates and mixed microbial cultures obtained in WP4 • enzymes obtained by the metagenomic approach of WP6 • screened by using standard substrates and micro-array mapping • testing assays will involve both 96-well colorimetric, and high-throughput HPLC, GC and Maldi TOF MS/MS • 1L stirred tank bioreactor under batch conditions (10 strains) • Ecotoxicity of the biosurfactants
Task 7.2 - Development of small scale bioprocesses for the reductive dehalogenation of chlorinated compounds • Develop biofilters for seawater contaminated with Cl-solvents • Chemical measurements • Ecotoxicityof the biosurfactants • Slurry bioreactors for the ex-situ removal of PCB • Biotransformation of PCBs • Testing assays will involve both 96-well colorimetric, and high-throughput HPLC, GC and Maldi TOF MS/MS
Task 7.3 - Development of novel biopalladium nanoparticles dechlorination processes • Optimize the production of bio-Pd using different isolates • Size & properties of the nanocatalysts • Optimize the biogenic hydrogen production • Larger scale reactor systems • - Cl-solvents analysis • - PCB analysis
Task 7.4 - Definition of a fate model for metal and metalloids in polluted sediments • Modeling of the fate of metals and metalloids • Based on sediments previously sampled and characterized in WP2 and WP3 • Biotreatmentexperiments • Data elaboration and ecological modelling of the bioremediation process
