  1. Mechanotransduction measurements in cells Modern instrumentation (phy 651) symposium Department of physics, Syracuse University EleniKinfeDegaga

  2. Introduction • Mechanotransduction? • How do the cells respond? • During differentiation how do the cells decide what they want to be? • Structure, dynamics and function

  3. The Membrane Skeleton of Cells Cell Membrane Cell Substrate/Environment Mechanical Forces between Cell and Substrate Ankyrin Spectrin Actin P55 4.1 βspectrin αspectrin

  4. Donald E. Ingber1 (2006),The FASEB Journal,20

  5. Khan, S., and Sheetz, M. P. (1997), Annu. Rev. Biochem. 66, 785–805

  6. Objective? - Measuring mechanical stimuli pressure, Force, Temperature, light intensity, humidity, And the biological component Strain, Elasticity, rigidity, stress, tension,Young's Modulus

  7. LabVIEW application to mechanotrunsduction • For atomization of mechanical stimulus application • Measuring the physical characteristics of the stimulus and the biological component • Simulation of possible mathematical models relating the mecanotransduction and the cell dynamics

  8. Strain measurement simulation • Required materials • LabVIEW 2009 containing - Control Design toolkit 2.1 -Simulation module 8.2 -Strain gauge and half bridge circuit -Biological organ sample

  9. Strain Guage

  10. Mathematical representation for strain model

  11. Mathematical model V1= [R3/(R3+R4)-R2/(R1+R2)] V2=[RG+changeinR/2RG-R2/(R1+R2)]Vex Strain= 2(V2-V1)/VEx(Gf)

  12. PID controller • Proportional, Integral and differential controlling • Proportional (Kp):- reduce rise time & steady state error • Integral (Ki):- Eliminates the steady state error, reduces the rise time, inreases settling time • Differential (Kd):- Increases the stability, reduce overshoot

  13. Strain simulation block diagram

  14. Show your program running for different experimental values
