Methode • Hot Treatment • Meristem Culture • Hot treatment, followed with meristem culture methode • Adventif root forming, followed with meristem culture • Meristem grafting on free viruses seed root (micro grafting)
Procedure • Identification of viruses existence and variety • Efforting to eliminate viruses • Examining of free viruses plant produced • Preventing of re-infection viruses
Free viruseswith Thermoterapy Hot air treatment Hot water treatment Spritus flame treatment
Hot air treatment • Principal: Placement the plant on therapy room and giving treatment temperature about 34-400C. • Long treatment, depend on species: • Pottao virus PVX : some month, temperature 350C, free viruses shoot tip to graft with health potato shoot, and cultivavted by in vitro
Hot water treatment • Grains (with hard skin ) • The infected viruses grains put in filter, then giving hot water (about 100 0C) • Washing grains with rather cool water • The grains are germinated • Shoot/embryo/endosperm using as explant
Spritus flame treatment • Treatment using spirtus flame • Cleaning the material (ex. Potato tuber) • The material into spirtus/alcohol • Burned on spirtus flame until the fire dead • Germination of material • Shoot growth free viruses, using as cloning material for invitro culture
Procedureto preventing of viruses re-infection Quak, 1966; Theiler, 1981 Placement of plant in green house or Free viruses vector room Preventing of infection sources and eliminating of viruses vector (insect, nematoda). Take care the equipment during plant growth Pots and substrate must free diseases . Take care the free viruses material by invitro. Continuining selection and examination
Free Viruses with Meristem Tip Culture • ‘There are viruses gradient from root to shoot/tip, on free viruses tobacco Meristematic shoot tip free viruses (length 100-500 µm; corn 2-5 cm)
