The role of inflammatory cytokines in preeclampsia
4. Hypothesis Inflammatory cytokine levels will be increased in the RUPP brain tissue in comparison to the control.
5. Methods Immunohistochemical methods
Rat brains with skull intact were taken at the 18th day of gestation from both control and RUPP treated adult female rats and were snap frozen.
Sections were taken at 50 microns with a cryostat and fixed to slides.
Tissues were stained for IL-1B, IL-10, and TNF-a proteins.
Tissues were examined by microscope to determine the presence of positive staining for each protein and the region of staining.
6. Rupp ModelReduced Uterine Placental Perfusion model Small silver clip is placed on the bifurication of the uterine horn.
Clip creates a reduced oxygen environment which creates characteristics similar to preeclampsia.

8. Cytokine expression for Tumor Necrosis Factor-alpha was decreased in the RUPP model at day 18 of gestation.
9. Cytokine expression for Interleukin 1ß was decreased in the RUPP model at day 18 of gestation.
10. Cytokine expression of Interleukin 10 was decreased in the RUPP model at day 18 of gestation.
11. Conclusion There is a decrease in cytokine levels at gestation day 18 for the RUPP model.
