Infectious Disease Research at Upstate Medical University SUNY REACH ID/EMERGING Pathogens Meeting June 20, 2013
Major Focus Areas Pathogens: Viruses • DNA viruses (HSV, VZV, CMV, EBV, KSHV, MHV-68) • RNA viruses (Dengue, Pneumovirus, RSV) Malaria Models: Model of Sepsis in Pigs Humanized SCID mouse models Antigen presentation using Listeria
Dengue virus • Epidemiology in Thailand and Ecuador (Endy, Polhemus, Stewart, and King) • Pathogenesis (Princiotta, Winslow, King and Endy) • Clinical trial for vaccine (Polhemus, Endy)
Pediatric Infectious Disease Research team at Upstate Medical University Joseph Domachowske MD ManikaSuryadevara MD Cynthia Bonville MS
Research Interests • Laboratory studies on the innate immune responses to respiratory viral infections • Translational studies in children with respiratory viral infections • Clinical vaccine trials in children
Laboratory studies on the innate immune responses to respiratory viral infections • Models of severe viral infection using the pneumonia virus of mice and Sendai virus • Severe illness mirrors human infant infection with respiratory syncytial and parainfluenza viruses • Goals • Identify innate immune responses that are associated with illness severity • Develop and test compounds that target and attenuate this robust inflammatory response
Translational studies in children with respiratory viral infections • Respiratory syncytial virus epidemiology • Longitudinal birth cohort study • Identification of upper respiratory tract biomarkers for infant RSV infection severity • Develop and test compounds that target and attenuate this robust inflammatory response • Documentation of the degree of neutrophil activation during infant RSV infection (ex vivo)
Clinical vaccine trials in children • Vaccine perceptions of routine immunizations in families of resource poor children in CNY (with NYSOCHD and Salvation Army) • Global perceptions of vaccine safety and efficacy by providers of pediatric care in the U.S., Central America, Asia and Australia (with American Academy of Pediatrics, and Shot@Life (GAVI, UNICEF, WHO) • Intranasal RSV vaccine (Medimmune) • Intranasal parainfluenza vaccine (MI) • Intranasal quadrivalent influenza vaccine (MI) • Inactivated quad influenza vaccine (GSK)
Herpesviruses • Longitudinal birth cohort based in Kenya • Primary EBV, KSHV and CMV infection in infants and impact of malaria on viral persistence (Rochford, Chan) • Virus-Cell interaction • Stat3 activation by KSHV (King) • Inhibition of monocyte apoptosis by CMV (Chan) • DNA replication of HSV (Hwang)
Humanized SCID Mouse Model Core • Interaction of varicella zoster virus (VZV) with human cells and tissues using skin and thymus that is implanted into SCID/beige mice. (Moffat) • Evaluation of anti-viral compounds (Moffat) • Evaluation of anti-malaria compounds (Rochford)
Host defense against Intracellular Pathogens • Ehrlichiamuris infection of mice and role of IgM memory B cells (Winslow) • TB (Winslow)
