  1. Supplementary Figure 3 e-g Supplementary Figure 3. Ki16425 is not toxic against RMS cells. Panel E and F - Images of cells that were incubated in the absence (vehicle) or presence of Ki16425. Panel E– cells incubated with medium supplemented with 10% FBS, Panel F– cells incubated in medium supplemented with 0.5% BSA. Representative images are shown. Panel G - Chemotaxisto LPA of RH30 cells pretreated for 1h with 10 μMKi16425. Results are presented as a percentage of cells migrating to LPA gradient as compared to control cells pretreated with vehicle only. Based on fact that chemotaxis assay was run for 24 hours, exposure of RMS cells to Ki16425 significantly affect responsiveness of RMS cells to LPA for at least 36 hours [24 hours (time of assay) + 12 hours (cells kept in culture before performing assay)]. The recovery of responsiveness to LPA gradient after 24 hours indicates transient but not toxic effect of Ki16425.
