Highlights • Identified a new kinase regulatory mechanism in ERBB2 kinase domain. aC-b4 loop functions as an intramolecular switch to control ERBB2 kinase activity and is involved oncogenic activation • Studies of ERBB2 lung cancer mutants revealed that oncogenic mutation leads to altered substrate specificity of the kinase. Transforming activity of mutants results from acquired substrate preference for the adaptor protein GAB1
Highlights • Discovered small serine/threonine kinase (SSTK; TSSK6) and found that the kinase is essential for male fertility • Developed a strategy for rapid evaluation of bioreactor process changes in the manufacturing of mannose-6-phosphorylated therapeutic enzymes, and characterized the impact of inducing agents on glycan phosphorylation
Highlights of Kula Jha’s Research • Identified and characterized TSSK6 activating cochaperone (TSACC): a novel HSP70-binding protein critical to HSP90-mediated activation of TSSK6 (SSTK) • Demonstrated that HSP90 functions to stabilize and activate the TSSKs, a family of protein kinases required for spermiogenesis and fertility • Future work will focus on (i) identification of physiological substrates for TSSK6 and (ii) role of HSP90 in spermiogenesis
