Proline • Amino acid + compatible osmolyte • Remain unchanged, H2O-soluble, no effect on other molecules • First plants response water stress: • decrease growth rate and turgor + prevention of feedback inhibition proline synthesis. (Crawford, 1989) • protects membranes and proteins against dangers of dehydration • Accumulation in cytosol leaves in salt tollerant plants, but also other plants during salt stress..
Hypothesis • Proline accumulates when salt concentration becomes higher
Method • Proline can be made visible by reaction with ninhydrine ( purple complex) • Extinction muesurement, ED 520 nm • Making standard line with known concentration • Calculation proline amount
Discussion • No standard measures, but clear accumulation • In every plant, proline accumulates, what’s the difference? Tollerant plants don’t begin at 0! • Excellent results! • Hypothese correct without doubt • Further questions: • Does proline keeps rising? • What kind of trendline between salt and proline? • What is the correlation with other data?
