“PCR is one of those inventions like the internet, once you have used it, you cannot quite understand how people managed before it existed”
Nobel Prize 1993 • Invented by Dr. Kary Mullis, • Research Scientist, Perkin Elmer Cetus Corporation in April 1983.
What is PCR? • A method of “amplifying” specific DNA sequences. • A test tube system for DNA replication that allows a “target” DNA sequence to be selectively amplified several million fold in just a few hours.
How Does PCR Work? • http://www.abpischools.org.uk/resources/poster-series/pcr/pcranim.asp
PCR Reaction: • Template DNA • Primers • dNTPs • Buffer • Taq DNA polymerase
PCR Applications • Endless.
Problems with PCR • To be discussed……. • Example: Bias • GC rich sequences tend to be preferentially amplified over AT rich sequences • Other sources of bias?
Discussion • Paper #1: Saiki et al. (Mullis) Science • Paper #2: Bruce et al. • Presented by Mary Beth • Paper #3: Abdulmajwood et al. • Paper #4: Wang and Wang
