To-Do Orders Safety Lab Upkeep Protocols Oracle Calendar Lab Logo Friday, 03 January 2020
Orders • http://hpgl.umd.edu/mediawiki-1.9.3/index.php/List • All items that you would like purchased need to be entered into the lab wiki • You may temporarily write items on the white boards, but they must eventually go in the wiki for me to place the order • To prevent confusion, forgotten orders, redundancy, and other issues, only items entered into the wiki will be considered for purchase • Order items in advance, before we run out! • It may take me several days to place the order, several more days for it to process and ship, and it may still be backordered • You need to include the item number and vendor if you are reordering an item • If it is a new item, please research it and post the best option or several options for me to review • If necessary, post several options and ask me to check prices • Always include item numbers • A list of reagents is located next to the white board in the lab and under the shelves of chemicals – refrigerated/frozen items that came from TIGR are not on this list – check this list before you ask me if we have something
Orders • To Order • Items that need to be ordered • Put the newest entry at the top of the list • Follow the format shown at the top of the page • Tentative • Items we are considering for purchase, but don't need immediately • Reminders, not order requests • Ordered • I move items here after the order has been placed • Will be updated with the order date, the expected ship date, backorder information, etc. so check for updates frequently
Safety • First Aid Kit – Under white board by RA room door • Let me know if you use/remove items from the kit – it has a minimum stock requirement • Fire Extinguisher – Under white board by RA room door • Fire Alarm & Closest Exit – Stairwell to the left, down the hall • Shower – Across from TC room, next to the lab door • Eye Wash – Next to Sink – Run periodically (once every week or two) • Chemical and Biological Spill Clean-Up Kits – We should assemble these ourselves • MSDS – In RA Room File Cabinet - “Equipment Manuals” Drawer
Safety • Safety Glasses, Goggles, and Face Shield • Use when boiling solutions on the bench, or working with caustic chemicals or irritants • Use when working with the UV transilluminator w/o the shield (transferring or cutting gels w/ UV on) • Ask me about their locations • Hearing Protection • Must be worn when working with the sonicator • 3 pairs of earmuffs have been ordered • Broken Glass Containers • Empty the small containers into the large box next to the big centrifuge and reuse the small boxes if possible • Chemical Waste Containers? • Wear Lab Coats!
Lab Upkeep • Drying Rack • Place washed glassware/plasticware on the drying rack, not on the bench surrounding the sink • When it is dry, return the glassware/plasticware you washed to the cabinets • Don't leave stirbars, forceps, or other small items near the sink – dry them with a papertowel and put them away • Sinks • Clean up after yourself and wipe down the sink at the end of the day to prevent staining, sitting water, etc. • Don't leave items in the sink – they can break or go down the drain • DO NOT WASH GEL SOLUTIONS DOWN THE SINK • Let larger volumes of gel solutions solidify (in the glassware, gel tray, etc...) and appropriately dispose of as solid waste (trash or EtBr waste) • If it is a very small amount, ensure it is a liquid and dilute with a large volume of water before washing it down the sink so it doesn't resolidify
Lab Upkeep • Common Equipment Areas • Proposal 1 – Everyone cleans up after themselves and returns the equipment to a state immediately usable by the next person • Proposal 2 – Each person is responsible for keeping the common equipment areas in their bench row clean and organized – I will maintain equipment not on lab bench rows, such as the balances and pH meter • Of course, tell me of any equipment malfunctions or problems • Hoods • Keep them clean and sterile • Remember to turn them off when not in use • There is a dust pan and broom by the broken glass container between the refrigerator and centrifuge • Trash needs to be put outside the lab at least weekly • Perhaps set the bags in the hall so we don't lose our trashcans
Protocols • http://hpgl.umd.edu/mediawiki-1.9.3/index.php/Protocols • No one has added any protocols to the wiki since I have on September 5th • Please write up your more permanent protocols using the format of an existing protocol as a template • It probably isn't worth generating a protocol for a method you are still trouble-shooting, but it would be a start • You may create separate pages or sections within Protocols to better organize them
Oracle Calendar • http://calendar.umd.edu/ • Schedule and Modify Lab Meetings • Post Important Dates and Seminars • Automatic E-mail Notifications • Accessible Anywhere • Need UIDs to Set Up Accounts • Log In Using Directory ID and password
Miscellaneous • Fyrite Kit • Try using Brenda’s fyrite kit – Camila • If we like it, we can share Brenda’s and purchase the fluids or we can buy our own kit • Biomek Robot • http://hpgl.umd.edu/mediawiki-1.9.3/index.php/Biomek_FX_Robot • Order extra supply containers for degassed pump water • Preliminary Projects • Automate PCR using an entire plate of primers • Automate PCR by picking primer pairs from plates of primers • Automate BP Clonase II Reaction to transform the PCR products into the Entry Vectors
Lab Logo • Lab Name • Host-Pathogen Genomics Laboratory • Can we be more unique? • T. brucei • Protein Interactome • T. cruzi • MASP
