This study aims to analyze the gene expression patterns of human hepatic stellate cell (HSC) during proliferation, identify critical genes involved in hepatic fibrosis, and understand the underlying mechanisms. The HSC cell line was treated with Platelet Derived Growth Factor (PDGF-BB) at various time points, and gene expression was assessed using microarray analysis. The results will contribute to our understanding of liver fibrogenesis.

Introduction • It is important to discover the mechanisms related to the proliferation and apoptosis of hepatic stellate cell line which has the critical role in liver fibrogensis. • Aim : Analyzing gene expression patterns of proliferation induced human hepatic stellate cell line (HSC) in time courses (0, 2, 4, 8, 12, 24, and 48 hr), and further identifying critical genes involved in hepatic fibrosis
Experimental Procedures • Cell line and treatment : Immortalized HSC with hTERT was treated with Platelet Derived Growth Factor (PDGF-BB) 10ng/ml for 0, 2, 4, 8, 12, 24, and 48 hr. Following treatment, cells were processed for total RNA isolation with Trizol. RNA quality was checked with Agilent 2100 Bioanalyzer (Agilent Technologies GmbH, Germany). • Microarray : Total 10 chips,19K Catholic human oligo chips from Microdissection Genomics Research Center of Catholic University, were used; four chips for internal controls (0hr), and one chip for 2, 4, 8, 12, 24, 48 hr each. Universal human reference RNA was used as the reference RNA. A two-color microarray hybridization method was used. The reference RNA was labeled with Cyanine-3, and experiment samples were labeled with Cyanine-5.
