WP6 presentation Quality of oats : protein and oil contents Micronutriments and avenins AVEQ meeting BUCHAREST 19-21/10/2010
Partners for this workpackage : France : Jean Koenig INRA Clermont-Ferrand Gérard Branlard INRA Clermont-Ferrand Audrey Didier INRA Clermont-Ferrand External service : Vetagro sup Agnès Piquet Estonia : Külli Annamaa Jogeva Plant Breeding Institute Romania : Danela Murariu Genebank Suceava Manuela Ibanescu Genebank Suceava AVEQ meeting BUCHAREST 19-21/10/2010
Preliminary work to analyses : Dehulling of seeds Done by Alf Seplitis (Partner 2) and Mathias Hermann (Partner 3) Hand dehulling has and will be done by IHAR Radzikov and PBI (Estonia) by hand which is a quite long work Thanks to them Milling has been done by laboratories carrying out NIRS analyses (problems of sticky flour..... ) AVEQ meeting BUCHAREST 19-21/10/2010
Near Infrared Spectra analysis (NIRS) On samples 2008 , have been realized for 6 locations : BUL, EST, FRA, ITA, ROM, POL, and on samples 2009 are on the way. Calibration by Kjeldahl (proteins) and (Fat) and Calibration verification have been done on 2008 samples. Calibration values will be entered in the NIRS software to produce protein and fat values. AVEQ meeting BUCHAREST 19-21/10/2010
Protein contentFirst results from calibration by Kjeldahl method show a wide range of variation : within locations and looking at all locations AVEQ meeting BUCHAREST 19-21/10/2010
Micronutriments • Have been realized on 122 (100 selected by PCA + 2 X 11 Standards) samples from 2008 (mineral micronutriments)on samples from french harvest. But results were not produced yet by partner (external) • Samples have been chosen on spectra from NIRS analysis (PCA) choosing more diverse accessions • Will be done on samples from France 2009 in the next months AVEQ meeting BUCHAREST 19-21/10/2010
Results for avenins patterns AVEQ meeting BUCHAREST 19-21/10/2010
Avenin identification Method • Avenin extraction • 1D electrophoresis Acide-Page • Allele identification for the 3 loci: Ave1, Ave2 et Ave3
Identification of alleles of Ave1locus • 5 alleles
Identification of alleles of Ave2locus • 6 alleles
Identification of alleles of Ave3locus • 10 alleles
Homogeneity (purety) of the Avena sativa patterns as revealed through avenin analysis • 22.4 % of accessions were heterogenous • Genetic heterogeneity within accession • Grain mixture within accession • 80% old accessions • 20% modern accessions
