Sysmex XE-2100 NRBC Detection
NRBC Counts - Methodology • From semiconductor laser flow cytometry. • STROMATOLYSER-NR (diluent) • STROMATOLYSER-NR dye (stain) • Passed through the laser flow cytometer for analysis. • Counted using side fluorescence and forward light scatter.
Flow Cytometry • Forward light scatter - cell size • Side fluorescence - RNA/DNA information (nucleus)
Resolution of NRBC and WBC • Clear & reliable separation of NRBC and WBC • Wbc take upp more dye than NRBC - so higher fluorescence • Smaller volume of the NRBC nucleus gives reduced forward light scatter.
The NRBC Count Algorithm NRBC NRBC% = NRBC# x 100 NRBC# + WBC# (NRBC/ 100 WBC) WBC# NRBC#
Eo Ly Ba NRBC Mo Ne The Corrected WBC Count WBC NRBC
NRBC # Revised Algorithm WBC region in WBC/Baso and Diff scattergram include NRBC if NRBC appear
WBC & DIFF channels: Correction for NRBC’s Correct WBC Minus Correct WBC Differential Minus
NRBC FlagsSwiching Algorithm NRBC-CH is measured “NRBC present” flag is monitored (WBC abnormal flag category) Detection from “NRBC%” NRBC-CH is not measured “NRBC?” flag is monitored (WBC suspect flag category) Detection from DIFF scattergram
