EXPRESSION OF WT1 AND CLAUDIN-5 IN ANGIOGENIC TUMORS S. Gobbo, A. Eccher, F. Comper, M. Brunelli, D. Segala, I. Cataldo, F. Menestrina, A. Scarpa, G. Martignoni 1Anatomia Patologica - Università di Verona
Background Identification of endothelial cells is based on the expression of angioblastic linage-specific genes like CD31, CD34. WT1: protein is a zinc finger transcription factor expressed in endothelial progenitor cells that has been reported to be maintained during the differentiation. Claudin-5: key component of tight endothelial junctions.
Methods Cases: 6 angiomas 5 Kaposi’s sarcomas 4 angiosarcomas Immunohistochemistry: WT1: anti-WT1, clone 6F-H2 from Dako diluted 1:100 Claudin-5: anti-claudin-5, clone 4C3C2 from Zymed diluted 1:100

Results: ANGIOMA WT1: consistent cytoplasmic and membranous immunoexpression claudin-5: expressed expecially on the luminal membrane with a week cytoplasmic labeling. WT1: strong cytoplasmic and membranous expression claudin-5: strong cytoplasmic and membranous expression WT1: strong immunoexpression in only 2 of 4 (50%) claudin-5: expression in the cytoplasm and on the membrane KAPOSI ANGIOSARCOMA
Conclusions: WT1 and claudin-5 are specific endothelial markers and are manteined in angiomas and Kaposi’s sarcomas. Angiosarcomas mantein the expression of claudin-5 whereas WT1, that has been proven to behave as a tumor supressor protein, tends to be lost in the development of the neoplasm.
