  1. FRAC SC - SDH inhibitors working group Frankfurt, 17. December 2008

  2. Already commercialised SDHIs

  3. amide group aromatic ring Common structural features of SDHIs Boscalid Carboxin Flutolanil Thifluzamid Penthiopyrad Furametpyr Source: BASF SE

  4. Carboxamides SDHI – Mode of Action:Inhibition of Succinate Dehydrogenase Inter membrane space Inner mitochondrial membrane C C C C

  5. All carboxamides have the same binding site Wrong!

  6. SDH A Fe-S cluster e- - transport ubiquinone binding site site of action of carboxamides SDH B SDH D SDH C Biochemical site of action: Structure of the SDH membrane

  7. Mechanism of resistance to Boscalid:Mutations in the target binding site H272Y,L,R,Q P225L,F,T Source: BASF SE

  8. Published and newer cases ofSDHI resistance in fungi

  9. Cross resistance between SDHIs • All SDHIs bind in a similar way in the ubiquinone-binding site • Cross resistance studies confirm that all SDHIs are affected by the mutations

  10. WG members

  11. Monitoring results 1/2

  12. Monitoring results 2/2

  13. WG meeting 5th December, outcome • Communication 1st step: Monitoring results • Telcon end January: exchange remaining data • Face-to-face meeting February: • Finalise guidelines for season 2009 • Discuss and agree on resistance risk statement* * Current status FRAC code list (#7): Resistance known for several fungal species in field populations and lab mutants. Target site mutations in sdh gene, e.g. H/Y (or H/L) at 257, 267, 272 or P225L. Medium risk. Resistance management required.

  14. Communication • 1st step (before end 2008) • Monitoring results • Background information on known target site resistance and MoA • List of known cases of SDHI resistance(Amendment of FRAC list) • 2nd step (1st quarter 2009) • Use guidelines for season 2009 • Resistance risk statement for FRAC code(including link to combined/ agronomic resistance risk assessment model)

  15. Thank you !
