Illuminator • Halogen Bulb • Usually controlled by a rheostat (knob that controls the intensity).
Condenser • Compound lens system that focuses the light from the illuminator onto the specimen. • Gathers a large amount of light and condenses it into a small area.
Aperture Diaphragm • Lever on the front of the condenser that allows you to control the amount of light leaving the condenser. • Opens and closes like the iris of you eye.
Stage • Simply the place where the slide sits. • This area becomes very dirty. Keep it clean.
Slide Holder • Mechanical device that holds the slide in place on the stage. Where ever the stage goes the slide goes!
Objectives • Traditionally come in four flavors in the lab • 10x, 40x, 50x, and 100x • Magnify the image • 50x and 100x require immersion oil. • The oil bends light waves so more light enters the objective
Oculars • Usually have a magnification power of 10x. • Form the final image. • Total Magnification: equals the oculars time the objectives. (40x times 10x = 400)
Focus Knobs • Two flavors of these: • Course • Fine • Some microscopes control the course and fine focus with one knob.
Stage Adjustment Knobs • Controls the stage movement. • Top knob controls forward and back movement. • Bottom knob controls left and right movement.
Cleaning the Microscope • Keeping the microscope clean is extremely important. • Use a clean, lint-free lens tissue. • Don’t use a dry tissue to clean the lenses. Use a moist tissue first and then a dry one to mop up. 4 3 5 2 1
Cleaning... • Start by cleaning the illuminator. • Move to the condenser • Next clean the objectives, but clean the 100x last because there may be oil on it. Once you have cleaned the 100x, get a new tissue. 4 3 5 2 1
Cleaning... • Clean the oculars • Lastly, wipe off the stage. Clean it last because it usually has the most dust and oil. • Make this a daily routine. 4 3 5 2 1
