A 2nd year college student working in the lab for the summer forgot to label the Tube of plasmid DNA that he/she was given. So being the brilliant graduate Student that you are, you digest the plasmid in the tube with 4 different enzymes. A picture of the stained agarose gel is on the left. See the handout for the sizes of the bands in the molecular weight ladder (MW). Is this plasmid in that tube? Start by determining the expected sizes of DNA fragments when each enzyme cuts the plasmid. Then using the molecular weight ladder, determine the sizes of the fragments in the gel. Compare.
What are the expected sizes of fragments if the plasmid above is cut with the following restriction enzymes: 1. BamH1 2. BglII 2. Bam H1 and BglII 3. NcoI 4. EcoRI 5. NcoI and EcoRI Compare your expected sizes to the gel on the next page. How do they compare?
