Quality of Lamellar Dissections Prepared with the 500 Khz Femtosecond Laser System • Purpose: • To study the quality of lamellar dissection prepared with the 500 kHz femtosecond laser (FL) • Materials and Methods: • 6 human globes were cut with the VisuMax 500 kHz FL. • 200, 300, and 400 micron deep cuts were made each at low (180 nJ) and high (340 nJ) energy settings [1.6 µ spot and line sep]. • Environmental scanning electron microscopy (eSEM) and atomic force microscopy (AFM) were performed on the tissue to assess the quality of the cuts.
Results Cut depth : 200 µm Energy : 180 nJ Cut depth : 200 µm Energy : 340 nJ eSEM • The 200 micron cut with high energy settings resulted in smooth dissections.

Conclusions • 200 and 300 micron lamellar cuts with the 500 kHz VisuMax FL result in excellent quality dissections. • “High frequency” roughness is lowest for the 300µm scans (both low and high energy). • Depth of cut and laser energy settings seem to affect the quality of the dissection.
