  1. DISCREPANCY IN NUCLEAR, MITOCHONDRIAL AND KARYOLOGICAL CHARACTERISTICS OF MUS MUSCULUS SUBSPECIES IN PRIMORYE HYBRID ZONE Spiridonova L.N. Institute of Biology and Soil Science, Far East Branch, Russian Academy of Sciences, Vladivostok 690022, Russia;spiridonova@biosoil.ru

  2. The aim of this investigations is: • Determination of congruence between taxon-specific markers on different inheritance systems (n-, mt-DNA and chromosomes) for revealing the pattern of real genetic structure of M. musculus in the extensive hybridization conditions in Primorye

  3. The tasks of investigation are: • To study subspecific structure of wild mice on nuclear DNA level in Primorye hybrid zone with RAPD analysis; • To sequence control region of mtDNA and identify subspecific haplotypes; • To compare molecular and karyological data for clear up real genetic structure of house mouse populations.

  4. The map of the collecting grounds 76 samples from 9 places of Primorye were used as the research material The wild mice were collected by colleagues of our lab (Korobitsyna К.V., Frisman L.V., Yakimenko L.V., Korablev V.P., Pavlenko M.V., Chelomina G.N.) and me.

  5. The primers used for studying

  6. RAPD-bands of M. musculus total DNA amplified with primer OPC-07 Vladivostok Kraskino Khasan Tury rog musculus castaneus domesticus 13 - gene GAS 1 (growth arrest specific 1) 1 - protein coding gene Aff3 17 - gene which is coding unknown hypothetic protein similar to retinitis pigmentosa 1 13 17 mus dom cas bac 1200 bp 1012 bp

  7. Phylogenetic tree constructed by ME method for M. musculus subspecies from sequences of mtDNA control region made with MEGA 4 m u s c u l u s castaneus domesticus outgroup

  8. The presence of M. musculus subspecific nuclear, mitochondrial DNA and chromosomes in Primorye mus* - own results, (R) – tandem repeat of control region segment from 67 to 81-bp mus(-)– from Yonekawa et al., 2003 ** - from Yakimenko et al., 2000

  9. The diagram of nuclear and mitochondrial subspecific characteristics of Mus musculus in Primorye musculus castaneus gansuensis domesticus

  10. The compared map of nuclear, mitochondrial and karyological data in M. musculus from Primorye mtDNA nDNA karyological data (Yakimenko et al., 2000) mus cas (dom?) gan Legend of nuclear and mitochondrial data 12, 15, 17, 18 C- C- C- C- C- 17 C± C± 10, 14, 15,18 C± 5, 17, 18, 19 C±

  11. Conclusions: • the inconsistency in markers distribution of different inheritance systems of wild mice was shown, that is the consequence of extensive introgressive hybridization in this region; • in most cases genetic diversity revealed by nDNA was higher than by mtDNA and chromosomes; • musculus mitochondrial haplotype (83%) predominates in all studied samples that fits to a pattern of absorption crossing; • the tendency for conservation of subspecific features of M. musculus (61%) is observed in the nuclear genome.
