  2. Topics

  3. Topics • Proficiency Tests • Chain of Custody • Methods • Membrane Filtration

  4. Proficiency Tests

  5. Proficiency Tests • Drinking Water: WS Study SW sample for MPN • Wastewater: DMR-QA WP Study • KY Approved PT Providers List

  10. Chain of Custody

  11. Chain of Custody • Client name & address • Sampler name • Sample location • Collection date & time • Sample type (grab/composite) • Matrix (DW/WW) • Field sample ID • Laboratory sample ID • Container type, & size/volume • Number of containers • Preservative (ice, etc.) • Field data (pH, TRC, etc.) • Requested analysis • Reason for rejection or • special remarks • Signature, date & time for • each transaction • (relinquishing & receiving) • Sample temperature upon arrival at lab

  13. Methods

  14. Microbiology Methods

  15. Colilert-18 P/A Colilert-18pre-warming

  16. Colilert-18 MPN

  17. Membrane Filtration

  18. Membrane Filtration Dilutions: Select appropriate volumes Fecal Coliform or E. coli colonies: 20-60* Total colonies: 200 *Use range as specified in particular method. Photo: www.pall.com

  19. Membrane Filtration Blanks: • Beginning of series • End of series • RECORD results of blanks

  20. Membrane Filtration Calculating Results: • Use dilution that is in range to determine result. • If multipleplateswithin range, average those plate counts. • If zero colonies on all plates, calculate as if 1 colony on filter from largest volume processed. Include remark “<“ in comments. Photos: www.avocetlab.com

  21. Membrane Filtration Calculating Results: • If all plates are above range, calculate using filter from smallest volume processed. Include remark “Estimated“ in comments. • WW TNTC = 60,000
