Safety in the Lab: • - Some of eye protection is required at all times. • - Never work alone • - Be familiar with physical properties of all chemicals used in the laboratory. • - Eating, drinking in the lab are prohibited • - Unauthorized experiments are not allowed • - Mouth suction should not be used
Cleaning Laboratory Glassware • Why to clean it?? - Any contamination could affect the results - Many chemical reactions are sensitive to one or more of the allowing: metal ions, detergents, organic residues.
Cleaning Laboratory Glassware • Cleaning Plastic ware: • Polyethylene containers: • when used for the 1st time, should be cleaned in 8 M urea ( adjusted to pH 1 ). After rinsing with Dis. H2O, wash with 1 N KOH, Dis. H2O again, then 10 -3 EDTA to remove metal ion contamination Finally rinse thoroughly with Dis. H2O
Cleaning Laboratory Glassware • Cleaning Glassware: • - washing with dilute detergent, (0.5 % Dis H2O) • If special washing is essential: • -Soaking in acid dichromate , and rinsing with Dis H2O (to free the glass of organic contaminants) • Second wash with conc. Nitric Acid, followed by Dis H2O (to remove metal ions) • Dichromic acid solution preparation: • Dissolve 5 gm of sodium dicchromate in 5 ml water in 250 ml beaker • Slowly add with constant stirring 100 ml conc. Sulforic acid • Temp. will rise to 80 C. allow solution to cool to 40 C and transferee it to dry bottle with glass stopper for storage. • Take care, as chromate is determined as carcinogenic!
Cleaning Laboratory Glassware Drying Plastic ware & Glassware: • For Glassware: Rinse with acetone which rapidly evaporate, then oven dry • Most plastic ware may also be dried in oven, but never dry cellulose nitrate centrifuge tube in an oven !! • Cellulose acetate is an explosive!!!!
Cleaning Quartz & Glass Cuvettes • - Never clean with ethanolic KOH or other strong base. • (it will cause etching) • A cuvette is a small tube of circular or square cross section, sealed at one end, made of plastic, glass, or fused quartz (for UV )and designed to hold sample for spectroscopic experiments. • A cuvette should be cleaned carefully with, Warm water with detergent, Dilute acid rinse, Copious dis.H2O rinse
