Fall 2011 Research Plan – Main Projects • Project 1: In vitro and in vivo magnetic nanoparticle (MNP) guidance in brain • Project 2: Intrathecal experiments with MNPs for targeted drug delivery • Project 3: Gaussmeter optimization for increased sensitivity
Project 1 Tasks – MNP Guidance in Brain • Conduct and analyze appropriate CED experiments. Different parameters will be tested for optimal conditions. • Experimental setup and protocol • Type, size, and concentration of MNPs • Magnetic field orientation and strength • Injection flow rates and catheter designs/size • Agarose gel concentration • Eliminate problematic variables from the CED infusions of MNPs. • Setup and begin animal experiments. • Characterize the MNPs using Zetasizer at Rush. • Use MRI at Rush to analyze MNP distributions in tissue and gel. • Look into using an electric field to drive MNPs through agarose gel.
Project 2 Tasks – Targeted IT Drug Delivery using MNPs • Synthesis of magnetite core with gold shell MNPs. • Characterization of MNPs using the ZetasizerNano at Rush. • Design and construction of different in vitro spinal cords/canals. • Calibration of pumps used in the IT experimental setups. • Run experiments using external magnetic field to control and target MNPs to specific regions along spinal cord. • Vary experimental parameters (such as flowrate, pulse rate, MNP concentration, external magnet strength and placement, etc) • Use different types of MNPs and/or ferrofluids • Visualize using both a digital camcorder and the angio machine
Plan for September • Eliminate problematic variables from the CED infusions of MNPs. • Characterize the MNPs using the ZetasizerNano at Rush. • Design, construct, and run appropriate IT experiments, as well as help advise Joe and Indu for these experiments. • Help Indu with MNP synthesis. • Conduct first MRI experiment at Rush. • Acquire animal brain tissue and start in vivo experiments.
