  1. Negative Stain / Capsule Stain • Stain background NOT bacteria • Stain and heat fixed treatment might distort shape • Use Bacillusmegaterium • DONOT follow procedures from manual • Use India ink (acidic (-)) / Repulsion with cell wall (-)

  2. Procedure: NOHEATFIXATION • Smear prep (DO NOT HEAT FIX) • Add a small drop of India ink • Place a cover slip • Press gently on top of cover slip using a brown paper • Add a drop of oil on the cover • Examine under the microscope

  3. EX. Capsule Stain • Polysaccharide / protein / glycoprotein • Waxy / mucoidal / slimy structure • Pathogenecity by • Resisting phagocytosis • Attachement to host tissues • Resisitant to antibiotics

  4. Have capsules Streptococcuspneumonae Bacillusanthracis Klebsiella Alkalogenesfaecalis Azotobacter NOHEATFIXATION
