Principles of Autoclave Operation • Steam penetrates objects in the autoclave • Condensation creates negative pressure and draws in additional steam • Moist heat kills microorganisms via coagulation of proteins (when cells containing coagulated protein cool, their cell walls burst, killing the microorganisms)
