Sunday, 20 June 2010 Future 2010-2013: Rules development Introduction of new methods Ustilago nuda on Hordeum vulgare (Nordic) Amalgamation of 3 methods for Linum At SHC, it was decided to align the three methods 7-007, 7-017, 7-018 in a single method to detect Alternaria linicola, Botrytis cinerea, and Colletotrichum lini Peer validation included 3  labs: Volcani (R Kohen), SNES (V Grimault) and Canadian Food Inspection Agency (S Brière) The parameters which were different in the three ISTA methods were compared: incubation T°, light during incubation and length of incubation were selected as parameters

  9. A pretest was carried out in one lab at SNES, to define the conditions to be tested for the peer validation. As new ISTA methods include the addition of 50 to 130 mg/L of Streptomycin, we decided to add this parameter for comparison: With/withoutantibiotics: Streptomycin 0; 50; 130 and 200 mg/L at 20°C dark Dark, Light/dark, dark/NUV Temperature: 20, 22°C Lots: 15% per pathogen, 4*100 seeds Results of pretest did not show significant difference between conditions

Sunday, 10 June 2010 Test plan: Peer validation 3 levels of contamination per pathogen (healthy, low, medium) 4*100 seeds per condition 5 Conditions : 1: Malt agar without streptomycin, 20°C, in darkness as 7-007 2: Malt-agar with streptomycin 50 mg/L, 20°C, in darkness during 9 days and 12h NUV/12h dark. 3: Malt agar without streptomycin, 22°C, 12h NUV/12h dark as 7-0017 and 7-0018 4: PDA with streptomycin 50 mg/L, 20°C, in darkness during 9 days and NUV 5: PDA with streptomycin 50 mg/L, 20°C, 12h light, 12h dark Evaluation 5, 7, 9 days
