Supplementary Fig. 1 Gated on lymphocytes TCR-gd CD161 SSC Total PBMC Gated on abT cells CD3 FSC 3C10 (Va7.2) Supplementary Fig. 1: Gating strategy for MAIT cells. PBMC were stained with mAbs against CD3, TCR-gd, CD161, and 3C10 (Va7.2), and gated on lymphocyte (left), CD3+TCR-gd- (middle), and then on 3C10+CD161hi (right) cells to identify MAIT cells as reported by Martin et al (11).
Supplementary Fig. 2 * * Supplementary Fig. 2: IFN-g production from PBMC depleted with MAIT cells in response to CD3 or CD3 and CD28 stimulation. PBMC with (DMAIT) or without (Total) depletion of MAIT cells were stimulated with plate-bound anti-CD3 mAb (5 mg/ml) alone (left) or in combination with soluble anti-CD28 mAb (1mg/ml) (right). IFN-g in the supernatant was measured at 48 hours. Results from 4 HC are shown. *p<0.05 (Paired t-test)
