  1. KEY CONCEPT DNA fingerprints identify people at the molecular level. DNA fingerprinting can identify people at the genetic level. And it allowed the child shown, called Baby 81 by rescue workers, to be reunited with his parents months after a tsunami, or tidal wave, devastated many parts of Southeast Asia.

A DNA fingerprint is a type of restriction map. • DNA fingerprints are based on parts of an individual’s DNA that can be used for identification. • based on noncoding regions of DNA • noncoding regions have repeating DNA sequences • number of repeats differs between people • banding pattern on a gel is a DNA fingerprint Infer: How would DNA fingerprints change if a different restriction enzyme cut the DNA in the middle of one of the repeated DNA sequences?

(mother) (child 1) (child 2) (father) DNA fingerprinting is used for identification. • DNA fingerprinting depends on the probability of a match. • Many people have the same number of repeats in a certain region of DNA. • The probability that two people share identical numbers of repeats in several locations is very small. Analyze: What is it about the DNA fingerprints that indicates each child is related to these parents? Fig. 3.1 - DNA fingerprints can be compared to identify people. Both children share some bands with each parent. Synthesize: Does a DNA fingerprint show a person’s genotype? Why or why not?

  4. 1 1 1 1 500 90 120 5,400,000 x x = = 1 chance in 5.4 million people Individual probabilities are multiplied to find the overall probability of two DNA fingerprints randomly matching. • Example: 1 in 500 people have 3 copies of the repeat at location A. 1 in every 90 people have 6 copies of the repeat at location B. 1 in every 120 people have 10 copies of the repeat at location C. Therefore, when the 3 separate probabilities are multiplied, suddenly the chance that 2 people have the same DNA fingerprint is very small. • Several regions of DNA are used to make DNA fingerprints. Most forensics cases use at least 5 regions of DNA. Summarize: How does identification by DNA fingerprinting depend on probability?
