Using Metabolomics to Investigate Implications of Diversity in Coral-Symbiodinium Unions Maggie Sogin Ph.D. Student Ruth Gates Lab
Not All Corals Are Created Equally • Differential coral response to GCC • Known diversity in Symbiodinium lineage • Macro-scale differences in physiology • Gap: what causes differences in physiology? 14C incorporation (dpm zoox-1 cm-2) DCMU treatment (µg l-1)
Hypotheses H1: The metabolome is different between different coral-Symbiodinium unions -How is the metabolome different? H2: The metabolome is different when photosynthesis pathways are turned on and when they are turned off. H3: The metabolome will change as a function of thermal stress from baseline measurements
My Methods Halt Metabolism Freeze Dry Over Night Crush Nubbins Removes water from tissues Coral Nubbin Liquid N2 Make CHCl3/ MeOH/H2O extracts Perform 1H-NMR Analysis Polar Non-Polar Process Spectra Multivariate Stats Match Spectra Data To Databases Principle Component 1 Principle Component 2
Current Progress 3d Stress: 0.06 • Collected samples from 4 corals at two time points (12 pm and 12 am) • Collected samples from P. lobata and S. hystrix after thermal stress experiment in Taiwan • Planning to present data at ICRS and WSN • Collecting additional data with samples for a manuscript Multidimensional Space Diagraph (MDS Plot) of Porites( ), Seriatopor ( ), Pocillopora( ) non-polar NMR data Principle Component Analysis from Porites lipid Profiles, light metabolome ( ) vs. dark metabolome ( )
