UF/FSU NMR/MRI News and Collaborative Efforts • Art Edison agreed to serve the NHMFL as Associate Director for Chemistry and Biochemistry • This summer he led an effort to write a proposal for a 950 MHz spectrometer focusing on 13C direct detection at high fields with both cryo and HTS probes - the latter to be jointly developed by Bill Brey and Agilent, building on the previous collaboration between Bill Brey and Rich Withers, formerly of Bruker. • Funding for a new HTS probe has been achieved represents a similar collaboration to that described above, but here for a higher mass sensitivity probe at 1.5 mm with enhanced filling factor. • The External Advisory Committee met in August and recommended that the vacant chief scientist position be filled with a Chem Bio researcher with major NIH funding.

FSU Facility Highlights from ‘08 • Installed Bruker Biospin Avance III console for the 900 and the Avance I 900 console will replace the current DRX console on the WB600 - this was part of a $3.2M contract for NMR instrumentation to FSU. For the 900 new console signal generation units and receivers, but with the previous RF power and gradient amplifiers as well as previous preamplifiers. Up to date software for the 900 and WB600. » state of the art capability for solids and imaging on the 900 and much improved capability for the WB600 - both WB 600 instruments will have Avance I consoles • New WB 500 - 50% for Prof. Anant Paravastu and 50% for the NMR User facility - primarily for MAS solids. A 1.3 mm 60kHz MAS probe will be delivered with the instrument
2009 Planned Proposals for NMR User Program Follow up on plans presented last year Proposal after 12 months of negotiations with NIBIB was refused for lack of interest in High Field Imaging and SSNMR. • Proposal to NIBIB for a BRP in support of solid state NMR and and Na+ imaging: “Novel Technology for Biomedical Research in Ultra-High Magnetic Fields” - letter requesting permission to submit proposal will be sent out next week. Proposal to be submitted 2/09. PI will be Cross • S10 Proposal to the NCRR ‘High-End NMR’ program for a 900 MHz console - to be submitted 1/09. PI will be Sam Grant Plan involved lease/purchase arrangement and University refused to permit us to submit it - FSU funded $2.5M and the NHMFL provided $0.7M for console upgrades. Proposal led by Art Edison for 950 MHz with 3 mm HTS probe submitted for High-End Instrumentation to NCRR • HTS 3mm probe for structural biology - to be submitted in late ‘09. PI will be Brüschweiler
2009 Planned Proposals for NMR User Program Follow up on plans presented last year • NIH proposal (mechanism undefined) for design/ construction of a 30 T high homogeneity HTS magnet - to be submitted in late ‘09 • NSF MRI proposal for the 36 T SCH console - to be submitted 1/10 After extensive deliberation it appears that we should obtain more preliminary HTS magnet results - 32 T HTS cold bore magnet for physics has been funded by NSF MRI. Proposal led by Bill Brey for SCH console submitted as an MRI Proposal to NSF Additional funding efforts • $9M Program Project Grant funded with UCSD, U. Alabama, Burnham Inst., & Harvard - validation of membrane protein drug targets - search for ligands that bind (initial drug screening) - structural characterization of 10 TB membrane proteins - development of triple resonance low-E probes for high fields - development of solid state NMR drug screening probes
Moving Forward - Continuing Facility Enhancement in Tallahassee - setting priorities • Last few years we pointed out that much of our instrumentation is seriously aging, some progress has been made. » UWB 900 - new console delivered in ‘09 » VNB 830 - needs a high power RF amplifier - will order Fall ‘09 » SB 720 - replace with a solution NMR centerpiece - 950 Proposal submitted » SB 600 - replace the 10 yr. old console - (along with 950 purchase) » WB 600a - an Avance I system - not currently a top priority » WB 600b - to be upgraded to Avance I in ‘09 » SB 500 w/cryo - receives little demand » (WB 500) - to be delivered in ‘09 » WB 400 - an Avance I console » SB 300 - staging instrument for the Keck magnet with TecMag console » WB 300 - important staging instrument - 14 year old DRX console » 36 T SCH - proposal submitted summer ‘09
Moving Forward - Continuing Facility Enhancement in Tallahassee - setting priorities - May ‘09 report to GSB Emergency Needs (list one item) Replacement of the 720 MHz instrument (950 MHz proposal - no resources if this is not funded) Top Priority Needs (list three items to maintain existing user program without growth) Spectroscopist scholar scientist for the 950 MHz instrument if funded Series Connected Hybrid Console if not funded (MRI R2 proposal - no resources if this is not funded) Expense and Equipment for RF development and maintenance Middle Priority Needs (list three items - necessary growth to maintain premier position) Jason Kitchen - RF engineer - temporarily funded on NMR account without budget Series Connected Hybrid Scholar Scientist Console for 600 MHz instrument (should be done in concert with 720 replacement) Emergency Needs (list five items - visionary growth in user program or strategic planning) Development of DNP Capabilities Animal Resource Technician (2 years of support from Grant’s start up) Software Application Engineer to facilitate software disbursement Replacement of two WB600 and 830 MHz consoles We would like to have your advice on these or other priorities
