Techniques included in training • Molecular biology techniques: • PCR ampilification and purification of PCR product • Mutagenesis methods (epPCR, Site Saturation Mutagenesis/Site Directed Mutagenesis) • Restriction cloning of Perhydrolase gene • Megawhop cloning • Transformation of E. coli and B. subtilis strains • Biochemical methods: • Expression of Perhydrolase in B. subtilis • Screening –determination of proteolytic activity and finding improved variants for oxidative stability • Purification of Perhydrolase • Electrophoresis techniques – SDS electrophoresis and Experion system
