SIMPLE STAIN (w/ Methylene blue)
Work in groups of two III. Gram Stain: Differential stain to distinguish between Gram(+) and Gram (-) Use the remaining smears of each bacterium you have already prepared Cover each smear w/ crystal violet for 1 min Grasp the slides w. a slide holder and rinse w/ water Cover the smears w/. iodine for 1 min Rinse w/ water Decolorize w/ ethanol for 2-3 seconds. ONE SLIDE AT A TIME Rinse w/ water Cover slides w/ the counter stain: safranin for 1 min Rinse w/ water Blot w/ bibulous paper, don't rub or wipe Observe under immersion oil Dispose everything in its place and clean Dispose extra stain inside the plastic bottle in the sinks
