(a) H3255 cells were transfected with either empty (Ctl) or p14ARF encoding vector. Expression of p14ARF, STAT3, pSTAT3-Y705 and Bcl-2 proteins was studied by western blotting after 6 days of G418 selection. Tubulin was used as a loading control. (b) H3255 cells were transiently transfected with control (Ctl) or p14ARF expression vector for 48 hours. The expression level of pSTAT3-Y705 was evaluated by pSTAT3-Y705 staining in p14ARF-transfected cells followed by FACS analysis. (c) H3255 cells were transfected with control or p14ARF siRNA. Western blotting was performed using indicated antibodies. Tubulin was used as a loading control. Long time exposure was performed for p14ARF immunoblotting in order to verify the efficiency of ARF neutralisation.
