For a Pass… • Safe working practice when carrying out paper and TLC. • Comment on suitabilityof technique (for both paper and TLC) • This standard chromatograms may not produce spots showing optimum degree of separation • Determine Rfvalues using paper chromatograms, using them to correctly identify components in a mixture.
A Merit… • Demonstrate safe working practices and a high level of proficiency when carrying out paper- and thin-layer chromatography (TLC) with minimal supervision. • Produce chromatograms showing clear separation of spots, repeating the separations if they are not satisfied with the quality of the separation obtained. • Comment on the suitability of the techniques for separation. • Use photographs to show how you carried out the experiment. • Use appropriately calculated Rf valuesand consider factors that influence separationto justify conclusions drawn about the identification of components in a mixture. • Polarity of components and solvents, effect of size of a molecule

A Merit Evaluate… ‘Quality of results obtained’ Repeats were done to get clear separation of spots.Why? Compare repeats of the paper chromatography. Streaking on 1st attempt: • Due to too concentrated sample spotted? • Wide spread spot? • When placed in the solvent it touched the side thus the solvent didn’t run to the top? How did you improve your experiment so this wouldn’t happen again?
How can I change this chromatogram? How could we get lysine to move up the TLC plate?
A Merit Why did we repeat carrying out chromatography using TLC after paper chromatography? Did this alternative chromatography technique improve your results? • Why and How?

How could we improve the experiment? How would you improve the intensity of colours of the plant pigments? How would you get clearer spots on the amino acid paper chromatography? Could the solvent be contaminated? What would contaminate it and how would this affect your results? How would you get clearer/more distinct separation between spots? Comparing your Rf values to that of literature/class? Do they match? Or show a similar separation?
