Analysis of dietary fibre - selected method Enzymatic-Chemical Uppsala Method (AOAC 994.13 or AACC 32-25) Gas chromatographic-colorimetric-gravimetric procedure • TDF = neutral sugar residues + uronic acids + Klason lignin • Principle • 1. Measurement of NSP as neutral sugar residues by GLC • (Englyst and Cummings procedure, 1988) • starch removal by specific enzymes • precipitation of soluble polymers by ethanol • hydrolysis of polysaccharides to monosaccharides with H2SO4 • reduction and derivatisation • 2. Measurement of uronic acid by colorimetry (Scottmethod, 1979) • 3. Measurement of Klason lignin by gravimetric method as the sulphuric acid resistant residue (Theander et al.,1995)

Analyses for starch and dietary fibre • Plans for the next four months: • Additional standard samples (53 samples of 11 cultivars from field experiment in Estonia) at present are analyzing for dietaryfibreand they shouldbe completed within 10 day from now. • Analyses for starch and TDF (NSP only) in 70 selected accessions from trial inPoland (2008) and France, Sweden and Italy (2009).
