Immunoblot analysis of the expression of polµ in stably transfected cells. Polµ Methods: Fifty µg of total cells extracts were separated on 10% SDS-PAGE and transferred to PVDF membrane. The membrane was blocked 1 h with 5% non fat dry milk diluted in TBS-T (0.5% tween 20) washed twice with TBS-T then incubated at room temperature for 1 hour with the polµ antibody (1/500) diluted in TBS-T. The membrane was washed twice with TBS-T and incubated with the secondary antibody (horseradish peroxidase (HRP)- conjugated anti-rabbit IgG) for 1 hour washed three times with TBS-T and revealed using the ECL system (Amersham).
