Kanker – Familiale kankers • kanker is het gevolg van somatische mutaties • de meeste kankers zijn sporadisch (toevallig) • borst- of eierstokkanker treft één vrouw op 10 • bij 10% is er een familiale geschiedenis van borst- of eierstokkanker • kanker zelf is niet erfelijk, sommigen erven een genetische voorbeschiktheid om kanker te ontwikkelen

  4. Borstkankergenen: BRCA1 en BRCA2 2 grote genen: BRCA1 (op chromosoom 17): 7.094 baseparen (1863 aminozuren) BRCA2 (op chromosoom 13): 10.390 baseparen (3418 aminozuren) Het onderzoek is complex en daarom voorbehouden aan familiale gevallen.

Indicaties voor mutatie-analyse • Goede selectie van families •  3 aangetasten (en sommigen < 50) • families met  2 BRCA +  1OVCA • families met  3 BRCA onder 50 j • 2 eerstegraadsverwanten met  2 CA onder 50 j • families met borst-en ovariumcarcinoom (BRCA1) • families met borstcarcinoom bij mannen (BRCA2) • borstkanker < 30j (ook bij unieke casus) • minimum 1 aangetaste persoon in leven
